Mass Spectrometry Analyst (Ref. 2024-06-AC/55)

The IEO Mass Spectrometry technological Unit is seeking a motivated, enthusiastic person to optimize sample preparation protocols, operate LC-MS equipment, and manage proteomics data analysis pipelines.

 

Key responsibilities

  • Closely collaborate with research groups to plan experiments, communicate results, and provide analytical support.
  • Introduce, train and supervise users to sample preparation, equipment operations and provide evaluation support.
  • Operate and maintain Unit equipment.
  • Reliably document all procedures along QC standards.

Requirements

  • Sc. or Ph.D. in Chemistry, Biochemistry, Biology or a similar field.
  • Documented expertise in common proteomic assays and MS-related bench techniques (Protein extraction, digestion and clean-up protocols).
  • Ability to perform basic analyses in common software tools (i.e MaxQuant, Perseus, PD, DIA-NN).
  • Expertise in routine operation, maintenance and troubleshooting of instruments .
  • Propensity to work in a collaborative environment.

The MS Technological Unit

The MS technological Unit at the Department of Experimental Oncology (https://www.research.ieo.it/research-and-technology/technological-units/mass-spectrometry-unit/). provides MS-based proteomics analysis to all IEO scientists, including protein identification, protein expression profiles by label-Free or label-based methods (SILAC, TMT), interaction proteomics (AP-MS, proximity labelling) and characterisation of post-translational modifications.

The unit is equipped with state-of-art of Orbitrap-Hybrid mass spectrometers. The instruments are operating in bottom-up workflows (DDA, DIA) and are coupled to a variety of nano-flow chromatography systems (nLC-1200, Neo Vanquish and Evosep-one). The unit also makes use of an 1680 HPLC (Agilent) for offline peptide fractionation (RP-HpH, SCX). The unit has also implemented Quantitative Proteomics Made Simple (QProMS), an R-based algorithm, search engine-independent data analysis and visualisation tool (https://bioserver.ieo.it/shiny/app/qproms).
